Gene identification and sequence analysis of Wolbachia in several common mites in citrus orchards.

Abstract: Polymerase chain reaction (PCR) was adopted to examine the wsp, ftsZ, and 16S rDNA of Wolbachia in Amblyseius orientalis, A. eharai, Euseius nicholsi, Neoseiulus barkeri, Panonychus citri, and Tetranychus harti from the citrus orchards in Nanchang, Jiujiang, and Ganzhou of Jiangxi Province. It was found that only T. harti from the citrus orchards in Nanchang was infected by Wolbachia, with the fragments length of Wolbachia wsp(Accession No. GQ162482), ftsZ (GQ162483), and 16S rDNA (GQ162484) being 593, 758, and 896 bp, respectively, while the other five species were uninfected. The Wolbachia in T. harti was very similar to the group Epo in subgroup B, based on the phylogenetic analysis of wsp gene.
