Abstract: Microsatellite DNA has been widely used as a good genetic marker in molecular ecology. This paper reviewed its application in molecular population biology, molecular environmental genetics, and molecular adaptation. The isolation of the loci is the basis of microsatellite DNA genetic analysis.Compared with the traditional isolation method by genomic library, some novel approaches carried out recently, such as enrichment library,PIMA and FIASCO,are more efficient and timesaving. The details and compare-analysis of these approaches were introduced, which would offer good choices for researches on molecular ecology.
